textThere is experimental evidence in Ba/F3 cell lines that Tyrosine-protein kinase JAK1 (Jak1) and Signal transducer and activator of transcription 1 (Stat1) associate with Interleukin-27 receptor alpha subunit (Il27ra, WSX-1).
Only Jak1 can co-precipitate with Il27ra, and be co-precipitated wit anti-Jak1 Ab.
When Il27ra protein is expressed as a GST fusion protein, when phosphorylated, can associate with Stat1 but not with other Signal transducers and activator of transcription (STATs). Moreover unphosphorylated form of GST-WSX-1 produced in BL21 cell line did not associate with any STATs. This fact was confirmed also in Ba/F3 cell lines expressing Il27ra protein.
In conclusion only Stat1 was revealed to interact with Il27ra (Takeda et al. 2003). This is a black box event because the exact mechanism related to the STAT1 and STAT3 binding has not been described. There is no data on which STAT is bound first or whether they are binded and phosphorylated simultaneously.
