Query author contributions in Reactome

Reactome depends on collaboration between our curation team and outside experts to assemble and peer-review its pathway modules. The integration of ORCID within Reactome enables us to meet a key challenge with authoring, curating and reviewing biological information by incentivizing and crediting the external experts that contribute their expertise and time to the Reactome curation process. More information is available at ORCID and Reactome.

If you have an ORCID ID that is not listed on this page, please forward this information to us and we will update your Reactome pathway records.

commentFUNCTION Involved in DNA nucleotide excision repair (NER). Initiates repair by binding to damaged sites with various affinities, depending on the photoproduct and the transcriptional state of the region. Required for UV-induced CHEK1 phosphorylation and the recruitment of CEP164 to cyclobutane pyrimidine dimmers (CPD), sites of DNA damage after UV irradiation (PubMed:19197159). During NER stimulates the 5'-3' helicase activity of XPD/ERCC2 and the DNA translocase activity of XPB/ERCC3 (PubMed:31253769). Connects XPD/ERCC2 and XPB/ERCC3 during NER, retaining DNA near the XPB/ERCC3 active site, and stabilizing the complex in a different conformation than in transcribing TFIIH (PubMed:31253769).SUBUNIT Interacts with GPN1. Interacts with RPA1 and RPA2; the interaction is direct and associates XPA with the RPA complex. Interacts (via N-terminus) with CEP164 upon UV irradiation. Interacts with HERC2. During NER binds XPD/ERCC2 and XPB/ERCC3 of transcription factor IIH (TFIIH), bridging the 2 proteins (PubMed:31253769).INTERACTION Expressed in various cell lines and in skin fibroblasts.PTM ATR-dependent phosphorylation of XPA at Ser-196 is important for cell survival in response to UV damage.PTM Ubiquitinated by HERC2 leading to degradation by the proteasome.DISEASE The disease is caused by variants affecting the gene represented in this entry.SIMILARITY Belongs to the XPA family.ONLINE INFORMATION Nature's flaws - Issue 142 of September 2012
