| text | IRAK1 mRNA was found to contain putative miR-146 target sites. IRAK1 activity was downregulated in human embryonic kidney cells (HEK293) when IRAK1 was co-expressed together with miR-146 [Taganov KD et al 2006; Nahid MA et al 2009]. Site-directed mutagenesis in the miR-146 target sequence of IRAK1 led to a complete abrogation of the negative effect of miR-146 on IRAK1 [Taganov KD et al 2006]. These data suggest that miR-146 has a negative effect on mRNA expression of IRAK-1, which is a critical signaling mediator shared by TLR4, TLR2/1/6 and TLR5 pathways. miR-146 family is composed of two members - miR-146a and miR-146b which are located on chromosomes 5 and 10 respectively. Several TLR ligands and pro-inflammatory mediators (LPS, PGN, Pam3CSK4, flagellin, TNFalpha and Il-1beta) induced miR-146 expression in human acute monocytic leukemia cell line (THP1). Moreover, miR-146 expression was driven by NF-kappa B factor and reached a plateau in ~8 h after LPS challenge [Taganov KD et al 2006; Nahid MA et al 2011]. |
