| text | The fibroblast growth factor (FGF) receptors belong to a family of transmembrane receptors that possess a cytoplasmic tyrosine kinase domain. Upon ligand binding, these receptors undergo dimerisation which initiates the activation of the intracellular tyrosine kinase domain. This leads to autophosphorylation of tyrosine residues in the receptor intracellular region resulting in the full activation of the FGFR complex. This recruits the adaptor protein STUMPS aka Dof aka Heartbroken, to its cytoplasmic C-terminal region and also the negative regulator RasGAP aka vacuolar peduncle (VAP), a Ras GTPase-activating protein. STUMPS is phosphorylated and provides a docking site for the SH2 domain of the phosphatase and adaptor protein Corkscrew (CSW). There are no binding motifs present on FGFR receptor for the adaptor protein Downstream of receptor kinase (DRK). However, CSW is phosphorylated by FGFR kinase domain and provides the necessary docking site for the SH2 domain of DRK. It recruits, via binding to one of its SH3 domains, the Ras guanine-nucleotide exchange factor, Son of Sevenless (SOS). SOS is involved in activating Ras (RAS85D) by catalysing GTP exchange (see Ras/Raf/ERK pathway). Activated GTP-bound RAS85D now triggers the kinase cascade involving Polehole (PHL) aka Raf1, Downstream of Raf1 (DSOR1), and Rolled (RL) which characterises the Ras/Raf/ERK pathway. FGF ligands bind to FGFR in only three different combinations. Ligand Branchless (BNL) only binds to receptor Breathless (BTL) while ligands Pyramus (PYR) and Thisbe (THS) only bind to receptor Heartless (HTL). The FGFR BTL and its FGF ligand BNL control tracheal branching in the embryo, mesoderm migration over male genital discs, and air sac formation in the larva. The preliminary function of the FGFR HTL and its FGF ligands PYR and THS, is to control mesoderm migration during gastrulation. Later in development, among other functions, HTL is also required for the differentiation of cells that form the heart and hindgut musculature. DRK protein is additionally involved in interactions with the regulatory plasma membrane protein Sprouty (STY). |
