textFANCI is monoubiquitinated on lysine 523 in an FA core-complex-dependent manner (Sims et al., 2007; Smogorzewska et al. 2007). FANCD2 and FANCI are mutually dependent on each other for their respective monoubiquitination. However, multiple phosphorylation of FANCI but not monoubiquitination is crucial for FANCD2 activation following DNA damage (Ishiai et al., 2008).
