commentFUNCTION Required for hearing (PubMed:30380417). Plays a role in maintaining cochlear stereocilia bundles that are involved in sound detection (PubMed:15489334, PubMed:35752427). Ensures the restriction of TPRN to the basal region of stereocilia in hair cells (PubMed:30380417).SUBUNIT Interacts with TPRN; the interaction restricts TPRN to the stereocilum basal region.SUBCELLULAR LOCATION Concentrated at the basal taper region of stereocilia in early postnatal development (PubMed:30380417). In adult hair cells, concentrated at the basal region but also detected along the stereocilium shaft (PubMed:30380417).TISSUE SPECIFICITY Expressed in sensory hair cells in the cochlea and vestibular organ.DISRUPTION PHENOTYPE Mice appear normal at postnatal week 2, but develop progressive hearing loss associated with early defects in orientation and organization of cochlear stereocilia bundles (Ref.3). They have normal vestibular function (Ref.3). Reducing Tprn expression corrects stereocilium morphological defects and partially restores hearing in Grxcr2 mutant mice (PubMed:35752427).SIMILARITY Belongs to the GRXCR2 family.CAUTION Although it shares high sequence similarity with GRXCR1, it does not contain a canonical glutaredoxin domain.
