Binding of PI3K to ALK is probably mediated by an adaptor protein that has not yet been identified (Bai et al, 2000; Slupianek et al, 2001; reviewed in Chiarle et al, 2008). Consistent with studies with the NPM-ALK fusion protein, recruitment of PI3K to ALK has also been demonstrated downstream of PTN activation, and phosphorylation of AKT is observed after ALK stimulation with MDK (Stoica et al, 2001; Stoica et al, 2002). In contrast, demonstration of PI3K activation downstream of ALKAL1 or 2 binding has not been demonstrated to date. |
