| text | After promoter clearance and transcriptional initiation, RNA Pol I enters the elongation stage in which the nascent rRNA transcript is extended (reviewed in Girbig et al, 2022; Comai, 2004). Elongation rate is affected by the TFIIS-like Pol I specific subunit RPA12 (Tafur et al, 2019; Scull et al, 2021), as well as other elongation factors such as PAF1C and DSIF, which also affect elongation by RNA Pol II (Wada et al, 1998; Schneider et al, 2006; Viktoroskaya et al, 2011; Zhang et al, 2009). The rRNA gene is simultaneously transcribed by multiple RNA Pol I enzymes assembled in tandem, forming what is referred to as a Miller tree (Neyer et al, 2016; Osheim et al, 2004; Miller and Beatty, 196). And although not indicated in this pathway, processing of the emerging rRNA occurs concurrently with transcription, and this processing is also believed to affect rRNA transcription elongation (Turowski et al, 2020). Finally, RNA Pol I elongation is regulated by epigenetic factors such as the B-WICH complex, which acetylates H3K9 at the rRNA gene to increase rRNA transcription (Sarshad et al, 2013; Shen et al, 2013), and CBX3, which binds K3K9me2 in the rRNA gene and has been shown to facilitate co-transcriptional processing of rRNA (Smallwood et al 2012). In contrast, binding of PHF6 to the UBTF transcriptional initiator has been shown to decrease rRNA transcription rate by decreasing UBTF protein levels in an unknown manner (Wang et al, 2012). |
