Negatively regulates chlorophyll biosynthesis and seed germination in the dark, and lightinduced degradation of PIF1 relieves this negative regulation to promote photomorphogenesis. Binds to the G-box motif (5'-CACGTG-3') found in many light-regulated promoters. Promotes the expression of SOM, and thus modulates responses to abscisic acid (ABA) and gibberellic acid (GA).ACTIVITY REGULATION DNA-binding ability is inhibited by PCH1 and PCHL to negatively regulate the expressions of its target genes.SUBUNIT Homodimer (PubMed:25944101). Interacts with the photoactivated conformer (Pfr) of phytochromes A and B, PHYA and PHYB. Also interacts with APRR1/TOC1. Binds to RGL2, RGA and FHY3 (via N-terminus). Associates to PTAC12/HMR/PAP5 which acts as a transcriptional coactivator (PubMed:25944101). Binds directly to PCH1 and PCHL; this interaction facilitates its association with phyB and its subsequent light-induced degradation (PubMed:32061894).INTERACTION Mainly expressed in leaves, stems and seedlings, and, to a lower extent, in fruits, flowers and roots.DEVELOPMENTAL STAGE Accumulates progressively in maturating seeds to reach a peak in dry seeds (PubMed:23708772). Slightly induced upon seed imbibition (PubMed:23708772).INDUCTION Repressed by red (R) and far red (FR) light treatments in a phyB- and phyA-dependent manner, and via a PTAC12/HMR/PAP5 regulated process (PubMed:15448264, PubMed:25944101). Down-regulated by abscisic acid (ABA) and auxin (IAA), but up-regulated by cold and heat (PubMed:23708772). Follows a free-running tenous circadian rhythm, with slightly higher levels during in the light phase (PubMed:23708772).PTM Phosphorylated at Thr-10, Thr-197, Ser-202, Ser-464, Ser-465, Ser-466 and Ser-469 by CK2 (PubMed:21330376). Phosphorylated and ubiquitinated after an exposure to light (especially red and far-red), in a phytochrome-dependent manner. Modified proteins undergo a proteasome-dependent degradation. Its stability and degradation plays a central role in photomorphogenesis of seedlings.DISRUPTION PHENOTYPE Plants overaccumulate free protochlorophyllide in the darkness and exhibit photooxidative damage (bleaching) in subsequent light, probably caused by the photosensitizing activity of this tetrapyrrole intermediate.MISCELLANEOUS May be due to a competing acceptor splice site.
